Rupert Murdoch's UK publishing businesses will move their headquarters from Wapping to The Place next to the Shard and London Bridge Station next summer.
The staff of News UK, Dow Jones and HarperCollins will be housed together for the first time and will begin relocating to the new site in the summer of 2014.
Wapping-based News UK comprises The Sun, The Times and The Sunday Times.
The HarperCollins book publishing business is currently based in Hammersmith.
"Our new London location in the vibrant borough of Southwark will allow us to realise one core objective as the new News UK – to work more closely and creatively, and leverage our collective resources," said Robert Thomson, chief executive of News Corp.
Cllr Peter John, leader of Southwark Council, said: "I am delighted that News Corp will be moving to such an iconic location in Southwark – bringing new jobs and opportunities to this part of London.
"The move underlines Southwark's growing reputation as a place to do business and endorses our work as a council to generate real economic growth."
The Place was designed by Shard architect Renzo Piano and developed by the same Qatari-backed consortium as the adjacent skyscraper.
